Pimobendan 5mg Oral tablet
For veterinary use only
Category: Vasodilators (Benzimidazole-Pyridazinone derivatives).
Form: Oral tablet
Compound: Each tablet contains 5 mg Pimobendan.
Target Animal: Dog.
Indication: Control and management of mild, intermediate and acute congestive heart failure due to Atrioventricular valve (AVVI) or Dilated cardiomyopathy (DCM).
Mode of Action: Pimobendan stimulates and increases the contraction strength of myocardial cells by two mechanisms. First, by increasing the sensitivity to calcium in the heart muscle myofilaments, and the other by inhibiting the enzyme phosphodiesterase (type 3) and as a result, increasing the amount of cyclic adenosine monophosphate. The effect of vasodilation by pimobendan also takes place through the inhibition of the phosphodiesterase-3 enzyme in the smooth muscles of arterial and venous vessels.
Pharmacokinetic: During an oxidative reaction, pimobendan is converted into its active metabolite and then bound with sulfate or glucuronic acid and is excreted mainly through feces.
The bioavailability of the product after oral administration is 60-63%, while administering the drug with food reduces this amount, so it is recommended to administer the drug one hour before taking food.
Pimobendan is distributed in body tissues immediately after administration. Its average binding rate with plasma protein is 93%. The elimination half-life of pimobendan from plasma is 0.4 ± 0.1 hours and the elimination half-life of its active metabolite is 2 ± 0.3 hours, which is due to the high clearance of 90 ± 19 ml/min/kg and the low average remaining time in plasma is 0.5 ± 0.1 hours.
Dosage and Administration: It’s to be prescribed daily with a dosage of 0.5 mg/kg and in 2 days with a dosage of 0.25 mg/kg with 12 hours apart.
Contraindication: In cases of hypertrophic cardiomyopathy, coarctations of aorta and other clinical instances in which cardiac output ratio is not sufficient enough due to anatomical or functional factors.
Cautions: The safety of Pimobendan’s consumption is not yet evaluated for puppies under 6 months of age, cases of congenital heart problems, diabetes insipidus or other critical metabolic diseases, dogs selected for mating and pregnant or lactating dogs. Also, safety of drug’s prescription for cases of non-symptomatic heart problems or other heart related diseases which have factors other than Atrioventricular valve or Dilated cardiomyopathy is not yet approved. Must be kept out of reach of children.
Side Effects: In rare instances there have been cases of tachycardia and nausea which is depended on drug’s dosage and will be resolved once the dosage is reduced. Also, there have been few instances of diarrhea, inappetence and numbness in which all these symptoms will disappear once drug’s consumption is terminated. Finally, there have been few reports of high mitral regurgitation in those dogs suffering from mitral valve failure and are undergoing long period treatment with Pimobendan.
Preservation: In a dry place, at room temperature (below 30°C) and away from light.
